What is the best method to relist Ebay items through Vendio. From Inventory or Unsold Closed Auctions? I have been advised several times by the Vendio staff to relist through Inventory, however, they have recently advised me to relist using the Unsold Post Sales category.

You need to relist from the postsale to get the ebay credit for the relist.
I think they are trying to get people to not keep so many items in the postsale just for relisting.
Why not just do what works best for you?

If it is not advisable to relist from Inventory, then, what is the purpose of the Inventory listings.

The listings don't stay in the post sale forever but they do in the inventory. They are also easier to find in the inventory with the search.
The other thing with the inventory is that you set the quantity you have and it will tell you if no more are available.

with in 30 days, I relist from the "closed" section, if it has been over 30 days or a item I haven't listed in awhile, I use inventory.
Hope that helps.

for a relist "credit" go from post sales, otherwise your perpetual inventory will not be correct.
